His SpinTrap/His SpinTrap Kit - Minipreps

His SpinTrap is a single-use spin column for purification of histidine-tagged proteins. For optimal convenience, use His SpinTrap Kit, which contains columns and buffers.
His SpinTrap columns
His SpinTrap™ is designed for efficient, small-scale purification of histidine-tagged proteins directly from clarified or unclarified cell lysates. The columns are also used for screening of large numbers of small-scale bacterial lysates, as well as optimization of purification conditions.
Purification of unclarified samples on His SpinTrap™ columns minimizes loss of target protein caused by manual operations such as sample precentrifugation, transfer to centrifugation tubes, and collecting supernatant. In addition, loading unclarified sample directly to His SpinTrap columns reduces sample preparation time, which minimizes degradation and oxidation of sensitive target proteins.
His SpinTrap columns are used together with a standard microcentrifuge and one purification run takes approximately 10 min. For optimal performance, use His SpinTrap together with buffers prepared using His Buffer Kit, see His Buffer Kit - Convenient, Pre-made Buffers.
The columns are prepacked with Ni Sepharose High Performance (see Ni Sepharose™ High Performance - High-performance Purification), which is precharged with nickel ions for capture of histidine-tagged proteins. Leakage of nickel ions from the Ni Sepharose High Performance medium is negligible.
His SpinTrap Kit

SDS-PAGE (ExcelGel SDS Gradient 8-18) of unclarified and clarified E. coli lysate containing GFP-(His)6 purified using His SpinTrap. Similar purity and recovery was observed from both the unclarified and clarified sample (lanes 4 and 5).
Purifying histidine-tagged proteins with His SpinTrap is a simple four-stage procedure that can be performed in 10 min using a microcentrifuge:
(1) After placing the column in a 2 ml microcentrifuge tube, equilibrate by adding binding buffer and centrifuge
(2) Add sample, centrifuge
(3) Wash with binding buffer, centrifuge
(4) Elute the target protein with elution by centrifugation
